חכמים מונים חמשה כלי נגינה בהם היו מנגנים הלויים: נבל, חליל, כינור, חצוצרה וצלצל. כלים אלו ומניינם נקבעו מפני שהנגינה המשובחת מתקבלת מההתאמה ומהיחס הנכון בין הכלים השונים ומספר המנגנים.

The Levites are also guards in the Temple. They had stations in twenty-one places in the courtyard and on the Temple Mount. This guarding is a positive commandment.

The role of the guards was not to protect the Temple, but to warn the unclean and strangers not to enter places that were forbidden to them. Also, this guarding was for the honor of God, just as in the king's palace the palace guards were for honor. In addition, the Levites were in charge of the gates and their job was to open and close the Temple gates.

בזמן הקרבת הקרבן יש מצוה לומר שירה וזה היה תפקידם של הלויים. בכל יום עמדו על דוכן הלויים לפחות שנים עשר לויים משוררים, והם שרו בזמן הקרבת קרבן התמיד. הלויים שרו גם בעת הקרבת עולות ציבור וקרבנות מוסף של מועדים – שבת, יום טוב וכד'. על הדוכן יכולים לעלות בנוסף ללויים גם ישראלים מיוחסים, לנגן בכלי הנגינה אך לא לשיר.
